SMITH, JOSEPH E., AND ERNEST BEUTLER. Methemoglobin formation and reduction in man and various animal species. Am. J. Physiol. 2 I o(2) : 34.7-350. 1 g66.-The rate of methemoglobin formation and reduction in man, goats, sheep, horse, cattle, and pigs was studied. Methemoglobin formation in the various species was studied by the addition of sodium nitrite to hemoglobin solutions, and methemoglobin reduction was carried out on nitrited washed erythrocytes using glucose and glucose plus methylene blue. It appears that the susceptibility to methemoglobin formation is related to the methemoglobin reduction rate in such a way that a rapid methemoglobin formation rate is offset by a rapid methemoglobin reduction rate.

Antioxidant effect of different vitamins on methemoglobin production: An in vitro study
Nitrite intoxication occurs frequently in ruminants and equines. The most common treatment of this disorder is administration of 1% methylene blue, although the use of some antioxidant agents e.g. vitamins and complementary treatment may also be useful. The aim of this study was to evaluate the in vitro antioxidative effects of some vitamins on methemoglobinemia induced by sodium nitrite. For t...
متن کاملFructation Induces Hemin Degradation in Methemoglobin
Under prolonged diabetic conditions, HbA1c is produced from normal hemoglobin (HbA0) through a non-enzymatic glycosylation or glycation, a process which enhances the hemoglobin susceptibility to be auto-oxidized to methemoglobin (metHb). Here we are reporting that the non-enzymatic reaction between fructose and metHb (metHb fructation) induces significant changes in the gl...
متن کاملInterspecies variability in propylene glycol dinitrate-induced methemoglobin formation.
Interspecies variability of propylene glycol dinitrate (PGDN)-induced methemoglobin formation was studied in vitro employing erythrocytes from four separate species. The net rate of methemoglobin formation was significantly different among species with dog greater than guinea pig greater than rat greater than or equal to human. This order of susceptibility was maintained in stroma-free hemolysa...
متن کاملComparison of the metabolism and toxicity of dapsone in rat, mouse and man.
The metabolism and toxicity of dapsone was compared in vitro and in vivo in rat, mouse and man. Metabolism was assessed by high-pressure liquid chromatography-mass spectrometry and methemoglobin formation has been used as a toxic endpoint. The greatest toxicity in vitro was seen in microsomes prepared from male Wistar rats (36.6 +/- 1.5% methemoglobin), although toxicity was also seen in micros...
متن کاملIdentification of Intracellular Sources Responsible for Endogenous Reactive Oxygen Species Formation
The endogenous reactive oxygen species ("ROS") formation is associated with many pathologic states such as inflammatory diseases, neurodegenerative diseases, brain and heart ischemic injuries, cancer, and aging. The purpose of this study was to investigate the endogenous sources for "ROS" formation in intact isolated rat hepatocytes, in particular, peroxisomal oxidases, monoamine oxidase, xanth...
